摘要
本文应用化学发光免疫测定法,以辣根过氧化物酶(HRP)标记抗体,采用夹心法,用鲁米诺-对碘苯酚-H_2O_2发光体系,测定0.0μU~30μU猪胰岛素,双对数标准曲线的相关系数为0.990,绝对检测限0.05μU,比国产胰岛素免放测定盒灵敏20倍。
Chemilum nescent Im-
munoassay (CLIA) has both high sensiti-
vity of chemiluminescent analysis and
specificity of immunoassay, while it can
get rid of the disadvantages of RIA, such
as radioactive hazard and high cost of
facilities. In this report, horseradish pe-
roxidase (HRP) was used as the label to
prepare HRP-Ab conjugate (Ab standing
for anti-porcine insulin guinea pig anti-
body) and polystyrene beads were used
to form the solid-phase Ab. Porcine ins-
ulin was determined with sandwich pro-
cedure and para-iodine phenol added to
the chemiluminescent system of luminol-
H_2O_2 (HRP) as a chemiluminescence en-
hancer to improve the determination se-
nsitivity of HRP. o. 03 μU-30 μU of insu-
lin were determined with a limit of
detection of 0.05 μU, indicating that this
method is 20 times more sensitive than
China made RIA kit for insulin.
