槲皮素对大鼠癫痫持续状态后海马XIAP mRNA与蛋白表达的影响 被引量:9

摘要 目的研究大鼠癫痫持续状态(SE)后海马组织XIAP的表达变化及槲皮素对其表达的影响。方法建立氯化锂-匹罗卡品致痫大鼠sE模型,应用免疫组化和RT-PCR方法检测XIAP与caspase-3蛋白以及XIAPmRNA的表达。结果海马CA3区XIAP蛋白在SE后2h(0.5503±0.0172)起逐渐增加,8h(0.6221±0.0238)达高峰,与对照组比较(0.1507±0.0165),差异有统计学意义(P〈0.01)。海马CA3区caspase-3活性蛋白在对照组未见明显表达,在SE后4—72h明显增多。与对照组比较,SE组海马XIAPmRNA表达水平在2~8h增加(P〈0.01)。与SE组比较,槲皮素组海马XIAPmRNA表达水平及CA3区XIAP蛋白表达在8h、24h高于SE组(P〈0.01),CA3区caspase-3活性蛋白表达在8h、24h、72h低于SE组(P〈0.01)。结论XIAP可能参与了SE后神经元凋亡的调控过程,槲皮素可以提高SE后海马神经元XIAP的表达。 Objective To investigate the expression of XIAP mRNA and protein in the rat hippocampus and quercetin treatment following status epilepticus (SE). Methods The lithium-pilocapine model of status epilepticus was established in SD rat. XIAP and activated caspase-3 protein were examined using immunohistochemistry. XIAP mRNA were ex- amined using semiquantitative RT-PCR. Results Compared with the control group, the expression of CA3 XIAP protein in the SE group was increased gradually since 2 h(0. 5503 ±0. 0172 vs 0. 1507 ±0. 0165, P 〈0. 01 ), and reached a peak at 8 h(0. 6221 ±0. 0238 vs 0. 1507 ±0. 0165, P 〈0. 01 ). The expression of CA3 activated caspase-3 protein were increased generally following SE. Compared with the control group, the XIAP mRNA levels revealed increase at 2 h, 4 h and 8 h (P 〈0. 01 ) in the SE group. Compared with the SE group, theexpression of XIAP mRNA and protein was increased at 8 h and 24 h (P 〈0. 01 ) in quercetin group, but the expression of CA3 activated caspase-3 protein revealed decrease in the quercetin group at 8 h, 24 h and 72 h ( P 〈 0. 01 ). Conclusions The XIAP are involved in the regulation of neuronal apoptosis after SE. Quercetin may increase the expression of XIAP in the rat hippocampus following SE.
