反相高效液相色谱法测定狗血浆中萘丁美酮代谢产物方法研究

摘要 本文报道利用反相高效液相色谱法测定萘丁美酮在狗血浆中的主要代谢产物—6-甲氧-2-萘乙酸的方法。采用2-(6-甲氧-2-萘)丙酸作为内标;甲醇?醋酸钠缓冲液(60:40)为流动相;采用ODS反相柱;用254nm波长的紫外检测器检测。实验结果表明:相关系数为0.9998,提取回收率为87-93%。本法具有快速、简便、灵敏的特点,适用于萘丁美酮的药代动力学研究。 The paper reports the determination of 6-methoxy-2-na- phthyl acetic acid (MNA), a major met- abolite of nabumetone in plasma of dog using reversed phase HPLC. 2-(6-metho- xy-2-naphthyl) propionic acid (MNP) is used as internal standard. Methanol in 0.05M/L sodium acetate buffer is used as mobile phase. The procedure involves extraction of MNA and MNP from dog plasma using n-hexane/ethyl acetate and evaporation of organic layer under a ge- ntle stream of nitrogen in a water bath. The residue of the solvent extract is dissolved in mobile phase and separated on ODS column. The eluates is detected by UV detector at 254nm. Thd retention time for MNA and MNT is 8.15 min and 10.50 min respectively. For MNA. abso- rbance is linear with concentration from 0.7-34 μg/ml and the limit of detedtion is 0.3 μg/ml in dog plasma. Extraction recovery is 87-93%. This method can be used in studing pharmacokinetics and bioavailability of nabumetone.
