参附芎泽注射液对压力超负荷大鼠血流动力学和细胞凋亡的影响 被引量:2

摘要 目的:探讨参附芎泽注射液对压力超负荷大鼠血流动力学和细胞凋亡的影响。方法:参照Ohkusa等介绍的方法复制腹主动脉缩窄大鼠模型。左心室插管术测定血流动力学指标;S-P免疫组化方法和图像分析技术检测心肌细胞凋亡相关基因Bcl-2和Bax的表达。结果:与假手术组相比,模型组左室内压力峰值(LVSP)、左心室内压最大下降速率(-LVdp/dtmax)及左心室内压最大上升速率(+LVdp/dtmax)下降;收缩压(SBP)、舒张压(DBP)、左室舒张末期压(LVEDP)升高;Bcl-2蛋白表达光密度值(OD值)减少、Bax蛋白表达OD值增多。参附芎泽注射液组与模型组相比,LVSP、±LVdp/dtmax升高,LVEDP、SBP、DBP降低;Bcl-2表达明显增多而Bax的表达减少,均有显著性差异(P<0.05或P<0.01)。与依那普利组相比无显著性差异。结论:参附芎泽注射液对压力超负荷大鼠血流动力学有明显的改善作用,抑制左心室心肌细胞凋亡,提高心功能。 Objective: To explore the effects of Shen Fu Xiong Ze Injection on hemodynamics and cardiac myoeyte apoptosis in experimental pressure-overloaded rats. Methods : A aortic band was placed in SD rats via replication of Ohkusa etc. We measured hemodynamics by putting the tube into the left ventricles. The protein expression of Bcl-2 and Bax gene were investigated by S-P immuno-histochemial stain and image analysis system. Results: In model groups, LVSP, -LVdp/dtmax +LVdp/dtmax decreased and SBP,DBP,LVEDP increased as compared with the sham-operation group; the protein expression of Bcl-2 gene was reduced and the protein expression of Bax gene inereased(P〈0.05).Compared with the model group, LVSP, + LVdp/dtmax in administration groups were higher and SBP, DBP, LVEDP were lower than those of the model group(P〈0.05); the OD value of Bcl-2 protein and the protein expression ralio of Bel-2/Bax increased and the OD value of Bax protein decreased significantly in administration groups. Conclusions : The present study suggested that Shen Fu Xiong Ze Injection could significantly improve hemodynamies and inhibit cardiac myocyte apoptosis.
