火麻仁油安全性评价及血清抗氧化功能初步研究 被引量:31

摘要 目的研究火麻仁油的急性毒性、遗传毒性和亚慢性毒性,同时初步研究火麻仁油对大鼠血清抗氧化能力的调节作用。方法采用急性毒性试验、小鼠骨髓细胞微核试验、Ames试验、小鼠精子畸形试验和大鼠90d喂养试验对火麻仁油的安全性进行评价,并对90d喂养试验大鼠血清抗氧化指标进行测定。结果火麻仁油对雌雄性大、小鼠经口LD50均大于每公斤体重21.5g,属无毒类。3项致突变实验均未显示出致突变性。大鼠90d喂养实验各项结果均未见明显毒性,未观察到有害作用剂量水平为每公斤体重10.0g。此外,经90d喂养,每公斤体重10.0g组雌、雄大鼠血清SOD、GSH-Px水平提高,MDA水平降低,与大豆油组和普通组相比均差异有统计学意义(P<0.05)。结论在试验条件下火麻仁油为无毒物,未显示有遗传毒性作用及亚慢性毒性;血清抗氧化指标检测结果提示火麻仁油具有一定抗氧化功能,值得进一步探讨。 Objective To research the acute,genotoxicity and subchronic oral toxicity of the hemp seed oil,and to study the antioxidant function in serum of hemp seed oil.Method Acute toxicity test,Ames test,micronucleus test of born marrow,sperm shape abnormality in mice and 90-day feeding test in rats were used to assess the safety of hemp seed oil,and antioxidant indexes in rat serum were also determined.Results The oral LD_ 50 of hemp seed oil in mice was more than 21.5 g/kg BW,so the hemp seed oil belonged to actual non-toxicity.The results of Ames test,micronucleus test of born marrow and sperm shape abnormality in mice were negative.No obvious adverse effects were observed at the dose level of 10.0 g/kg BW in the 90-day feeding test in rats.SOD and GSH-Px levels increased and MDA level decreased significantly in rat serum at the end of 90-day feeding with hemp seed oil,in comparition with control groups of soybean oil and the normal diet,and there were significant differences in hemp seed oil group(P〈0.05).Conclusion No adverse effects were observed in the series of toxicity tests and hemp seed oil could be safe to eat at the level of 10.0 g/kg BW.The change of antioxidant indexes in rat serum indicated that the antioxidant function of hemp seed oil should be further evaluated.
