人类β1,3半乳糖基转移酶7对人4IgB7-H3的修饰作用初探

摘要 目的:探讨人4IgB7-H3基因转染人胃癌细胞株SGC7901后人类β1,3半乳糖基转移酶7(β1,3-galacto-syltransferase 7,β3GalT7)的表达变化,以期发现4IgB7-H3与β3GalT7的相互关系。方法:脂质体介导重组逆转录病毒载体pEGZ-Term/4IgB7-H3转染SGC7901细胞,筛选获得稳定表达4IgB7-H3的亚克隆细胞株,通过RT-PCR和蛋白质印迹方法检测β3GalT7的表达。结果:成功获得稳定表达4IgB7-H3的亚克隆细胞株SGC7901/4IgB7-H3;β3GalT7随4IgB7-H3的表达上调而上调。结论:4IgB7-H3蛋白的修饰成熟可能需要β3GalT7的作用。 Objective: We researched the change of UDP-Gal:betaGlcNAc beta 1,3-galactosyltransferase polypeptide 7 expression in SGC7901/4IgB7-H3 cell to find the relation between 4IgB7-H3 and 133GalT7. Methods: Transfered pEGZ-Term/4IgB7-H3 into SGC7901 cell, then detected the expression of UDP-Gal:betaGlcNAc beta i ,3-galactosyltransferase polypeptide 7 with RT-PCR and Western-blotting. Results: SGC7901/4IgB7-H3 cell line were successfully obtained. The expression of β3GalT7 was upregulated follow the up-regulation of 4IgB7-H3 on mRNA and protein level. Conclusion: UDP-Gal:beta- GlcNAc beta 1,3-galactosyhransferase polypeptide 7 may modify 4IgB7-H3 protein.
