Establishment of Genetic Transformation System in Chinese Cabbage

摘要 系统研究了根癌农杆菌LBA4404介导的大白菜遗传转化的若干因素,成功地建立了大白菜快速有效的遗传转化体系,并获得了大白菜抗性植株。实验结果表明,大白菜的不同基因型、苗龄、外植体类型,菌液浓度、浸染时间,共培养和抗生素浓度对转化频率均有一定影响,其最高转化频率达4.0%。抗性植株经PCR和Southern blot检测表明,目的基因已整合进大白菜基因组中。 Several factors affecting tissue culture and genetic transformation of Agrobacterium tumefaciens-mediated Chinese cabbage were studied, and a quick and effective protocol with optimized condition for genetic transformation of Chinese cabbage was developed. Some transgenetic plants have been obtained. Different gene types and explants of Chinese cabbage, bacterial concentration, infection time, and the co-cultivation media, the concentration of antibiotics have some effects on transformation frequency. The highest transformation frequency was 4.0%. PCR and Southern blotting analysis showed that the target gene has been integrated into the genome.
