创伤反应性神经体液ET_1表达与CNS超微结构实验研究

摘要 为了进一步探索局部肢体创伤中枢神经系统(CNS)应变内环境紊乱的某些病理生理基础,揭示了大脑应激受损的可能作用机理,为创伤后反应性神经、精神异常等CNS反应的研究提供一定的实验理论依据,本研究借助犬双后肢低、高速投射物仿应激模型,采用被免分析、RNA斑点杂交、计算机图像分析等技术手段,对内皮素-1(ET1)在神经体液中的表达与释放进行动态观察和定量分析,结合CNS超微结构早期病理形态学观察,深入探讨了CNS应变效应。结果发现,低速组脑脊浚(CSF)、血浆ET1伤后6h,高速组下丘脑、海马区ET1-mRNA表达增强(P<0.05),ET1释放增多((P<0.05);病理形态学观察仍以高速组下丘脑、海马区明显,主要表现为神经纤维微等微丝排列紊乱,部分纤维肿胀、层面分离现象及神经纤维均匀一致性改变,髓鞘脱失.结果表明局部肢体严重创伤,虽未直接位及大脑,但对CNS确有不同范围、不同程度的间接损害作用;且不同应激强度对CNS产生不同程度的影响.提示下丘脑、海马区选择性易损,可能为反应性CNS受损的病理生理基础之一。 The general purpose of the study was to research some pathophysiological basis of central nervous system (CNS) in disturbance of internal environment by extredhes wound,reveal the possible mechanism of stress damage of brain, provide an experimental theoretical basis for the studying of injury reactive neuropsychic disorder. On the basis of the stress mode of different veocity sed bullet injury to hind thighs in dogs, the radioimmunoassay, RNA dot blot hybridization and computer image pattern analysis were used to dynantically observe and quantitahvely analyse the expression of neurohwnoral endothelin 1(ET1),combined with observing the early pathomorphism chapes of CNS ultrastructure toprobe into CNS stress reachon responding to different stress intensity. The results showed that ET1 levels in CSF and plasm after b hours posttrauma in low sped group, ther 2 hours posttrauma in high speed group markedly increased (P<0.01), that the exprssions of ET1+mRNA in hypothalamus in low speed group, hypothalamus and hippocampus formation in high speed group wer sighficantly enhanced (P <0.05),and that the early pathomorphism changes displayed the stratiform dissociation and homogenous degeneration of some fibers in hypothalamus and hippocampus formation in high speed group. It suggests that in the condition of extremities injury, CNS may be indirectiy damaged in different range and level, and caused various stress reaction. It also suggests that thethalamus and hippocampus formation were vulnerably damaged in sgite of no direcr injury to head,and these may play a consistent pathophysiological role in the development of CNS irritable injury.
