摘要
目的:观察丹皮酚影响下离体成纤维细胞的增殖情况,为预防因成纤维细胞增殖引起的一系列病理过程提供理论依据。方法:取10只健康Wistar大鼠用于培养成纤维细胞,将离体大鼠腹膜成纤维细胞平均分为4组,分别加入细胞培养液RPMI-1640,RPMI-1640+TGF-β,RPMI-1640+丹皮酚,RPMI-1640+TGF-β+丹皮酚。测定各培养液中成纤维细胞增殖情况,并进行比较。结果:单独使用丹皮酚不影响成纤维细胞的增殖(P>0.05),单独使用TGF-β可以刺激成纤维细胞活化进入增殖状态(P<0.01),加入丹皮酚的干预后成纤维细胞的增殖率明显下降(P<0.01)。结论:TGF-β可以促进成纤维细胞活化,丹皮酚有抑制活化的成纤维细胞增殖的作用,应用丹皮酚可能在一定程度上减轻病理状态下成纤维细胞对人体的损害。
Objective: To study the effect of paeonol on the proliferation rate in fibroblast cells. Methods:Ten healthy Wistar rats were used to cultivate fibroblast cells. Cultured cells were divided into four groups,group 1 (RPMI-1640),group 2 (RPMI-1640+transforming growth factors-β,TGF-β),group 3 (RPMI-1640+paeonol ),group 4 (RPMI-1640+ TGF-β+paeonol). The proliferation rate was detected with MTT. Results: There was no change for proliferation rate in paeonol group. The fibroblast proliferation was motivated by TGF-β. The fibroblast proliferation was significantly reduced by paeonol and TGF-β. Conclusion: TGF-β can stimulate fibroblast cell proliferation,but paeonol inhibited the proliferation. It is suggested that paeonol might,in certain exrtent,reduce the impairment of fibroblast to human being under pathological state.
