实时荧光定量PCR检测MMP-3 mRNA在大肠癌的表达

摘要 目的探讨大肠癌组织MMP-3 mRNA表达及其临床意义。方法用实时荧光定量PCR检测56例大肠癌组织及癌旁正常组织MMP-3 mRNA表达,分析MMP-3 mRNA表达与大肠癌临床病理因素之间的关系。结果癌组织和癌旁正常组织MMP-3 mRNA表达有显著差异(P<0.05)。癌组织MMP-3 mRNA表达与淋巴结转移、组织学类型、血管侵犯、肝转移及肿瘤病理分期密切相关(P<0.05),与肿瘤部位、肿瘤大小、及患者的性别和年龄等无关(P>0.05)。结论MMP-3高表达同大肠癌生物学行为密切相关,可能与大肠癌的发生、发展及预后有关联。 Objective In this study,we investigate cancerous tissue MMP -3 mRNA expression in patients with colorectal carcinoma and evaluate its clinic significance. Methods FQ - PCR method was used to detect tissue MMP -3 mRNA expression in 56 patients with colorectal carcinoma and its relationship with many pathologic parameters. Results Tissue MMP- 3 mRNA expression was found to be significantly elevated in patients with colorectal carcinoma, compared with normal tissue ( P 〈 0.05 ). There was a close relationship between tissue MMP - 3 mRNA expression and the pathologic parameters such as presence of venous invasion, lymph node metastasis, histological type, liver metastasis and clinical -pathological stage (Dukes) (P 〈0.05 ) ,but its relation to tumor site,age and sex was not significant(P 〉 0. 05). Conclusions Tissue MMP- 3 mRNA expression correlated significantly with biological behavior of colorectal carcinoma, the high expression of MMP -3 might be related with the occurrence and progress of colorectal cancer.
