摘要
用反相高效液相法,对血浆非那西汀药物浓度的测定方法及其药代动力学进行了研究。实验采用Waters公司HPLC系统,国产ZORRAXBPCN柱,以磷酸盐缓冲液/乙氰为流动相,内标法定量。本方法标准血浆线性关系良好(r=09999),线性范围070~8928μmol/L,最低检测量为125ng,平均方法回收率为9992%,浓度为8928μmol/L、1116μmol/L的提取回收率分别为948%、831%和824%;精密度考察批内变异系数(CV%)分别为049、365和490,批间变异系数(CV%)分别为393、364和593。
A sensitive and precise RP HPLC method for determination of plasma phenacetin, a marker of cytochrome P 450 IA 2,and for its pharmacokinetical study was developed. Waters HPLC system, home made ZORRAX BP CN column and phosphate buffer/acetonitrile as a mobile were used.Methodological evaluation showed that plasma phenacetin concentration between 0 70 and 89 28 μmol/L had a good linear confficient relation (r=0 9999). The lowest detectable concentration and amount were 0 70 μmol/L and 1 25 ng respectively.The average recovery of the method was 99 92%,and the extraction recovery for the concentration of 89 29 μmol/L,11 16 μmol/L and 2 79 μmol/L was 94 8%,83 1% and 82 4% respectively and the average coefficient variation of within run and between run was 3 07% and 4 50% respectively.
