线粒体肌酸激酶同工酶在肺癌和食管癌组织中的表达 被引量:1

Expression of Mitochondrial Creatine Kinase in Human Lung Carcinoma and Esophageal Carcinoma

摘要 应用自制的兔抗人心肌线粒体肌酸激酶同工酶(CK-mimi)抗血清,采用ABC法对106例肺癌、52例食管癌存档蜡块进行了免疫组化定位检测。结果显示:肺癌CK-mimi的检出率为100.0%,阳性染色的肺癌细胞浆内可见清晰的棕色颗粒,以鳞癌细胞最明显,52例食管癌也全部为阳性表达。CK-mimi的染色强度随着肿瘤细胞分化程度的升高而减低,鳞癌的阳性染色最明显,腺癌中阳性颗粒细小,分布均匀。Ⅲ级食管鳞癌中CK-mimi的表达低于Ⅰ级和Ⅱ级食管鳞癌,有显著性差异(χ2=5.10,P<0.05和χ2=7.19,P<0.01)。表明细胞内CK-mimi的阳性表达强度和阳性颗粒分布与细胞的分化程度和病理分型有关,它可作为一项肿瘤标志物用于诊断恶性肿瘤,对肺癌、食管癌的防治研究具有一定的临床意义。 An immunohistochemical assay was carried out in paraffin sections of 106 cases of lung carcinoma and 53 cases of esophageal carcinoma by ABC method using rabbit serum antibody against mitochondrial creatine kinase isoenzyme (CK mimi) of human heart. The results showed that the positive rate of CK mimi in specimens of lung carcinoma and esophageal carcinoma were 100.0% In cytoplasm of positive staining lung carcinoma can be seen obvious brown granules, especially in cytoplasm of squamous cell carcinoma. Positive expression was also revealed in all 52 cases of esophageal carcinoma.The staining intensity of CK mimi was decreasing with increase in degrees of cell differentiation of the tumors. The positive staining in squamous carcinoma was most obvious and in adenocarcinoma the positive granules were tiny and evenly distributed. The expression of CK mimi in Ⅲ grade esophageal squamous cell carcinoma were lower than in cases with Ⅰ grade and Ⅱ grade esophageal squamous cell carcinoma( P < 0.05 , P <0.01).The results suggest that the positive granules of CK mimi may have some relationship with the histological types and differentiation and may be used as a tumor marker for the diagnosis of this malignancy. It has certain significance for study on prevention and treatment of lung carcinoma and esophageal carcinoma.
