Effect of Different Factors on Primary Culture of Carnation

研究了不同因素对香石竹初代培养的影响,结果表明:用75%酒精灭菌1min,再用0.1%HgCl2灭菌20 min后,香石竹外植体的成活率最高;以4号培养基对叶片进行愈伤组织诱导,其诱导率最高,达到了95%;以黄色香石竹叶片为外植体进行初代培养,诱导率最高,为88%。

The leaves of carnation were used as explants and effects the different factors on primary culture of carnation were studied. The results showed that 1 min with 75% ethyl alcohol and 20 min with 0. 1% HgC12 were the best method to reduce pollutions, and the carnation explants survival percentage was higher; the No. 4 medium was used to induce callus tissue, the inductivity was the highest and up to 95% ; the yellow leaves of carnation were used as explants and primarily cultured, the inductivity was the highest and up to 88%.
