A broad-suitable method for total nucleic acid extraction from plants and animals

摘要 依据最简单的核酸提取3步骤:细胞裂解、去除杂质、沉淀核酸的原理设计了一种普适性的动植物核酸提取方法(简称PS法),并对包括裸子植物门的银杏纲、苏铁纲、松柏纲,被子植物门的双子叶植物纲和单子叶植物纲的20种植物材料以及动物界的5纲5种材料进行了提取试验.结果表明,用PS方法提取动植物总核酸均能取得较理想的提取效果.所提取的总核酸分别用RNase和DNase进行消化,可分离纯化出高纯度的DNA和RNA. According to simple three steps for nucleic acid extraction, in cluding cells rtipturing, impurities removing and nucleic acid precipitation a new nucleic acid extraction method called PS method was worked out. Better results were obtained on the Ginkgopsida, Cycadinae, Coniferae, dicots and basis of extractions from 20 species of plants including monocotyledon of angiosperms, and 5 species of animals of 5 classes. High purity of DNA and RNA were obtained after digestion with DNase and RNase respectively. The extracted DNA and RNA were excellent for RT-PCR and SSR amplifications. The experimental results showed that the PS method was a broad-suitable method for nucleic acid extraction from plants and animals.
