摘要
用过滤面积为22cm2的过滤装置进行试验。研究中用721型分光光度计在600nm处测定糖化酶的透光率来衡量滤后糖化酶液澄清程度,运用正交试验确定了硅藻土助滤糖化酶液的最佳预涂量、添加量、粗细比,并对最大过滤量、硅藻土对酶活力的影响进行了研究。试验最佳预涂条件:预涂第1层粗土706g/m2,第2层细土188g/m2,第3层粗土706g/m2;添加量为20g/m2,粗细比为5∶2。
The filtration of glucoamylase fluid is tested with a filtrating apparatus of 22 cm2 in area. The light penetrability of this fluid is assessed by 721 spectrophotometer with the transparency of the filtered glucoamylase fluid at 600 nm. Optimum condition of precoating and the crudeness /fineness ratio of particles are determined by orthogonal experiment: the precoating of the first and third layer are 706 g/m2 (crude particles) and that of the second layer is 188 g/m2 (fine particles), the quantity of addition is 20 g/L, the crudeness /fineness ratio of particles is 5∶2.
