摘要
在酶法合成L丙氨酸过程中,产物L丙氨酸对酶有抑制作用。本文选用PFR型填充床式反应器,以k卡拉胶为包埋材料,包埋PseudomonasdacunhaeDQp1,在底物为1mol/LL天冬氨酸,反应条件37℃、pH6.0时,连续生产L丙氨酸。在转化率达到95%时,计算出固定化细胞和反应器的效率分别为0.015gL丙氨酸/g固定化细胞·h、0.013gL丙氨酸/ml反应器·h。还建立了填充床反应器停留时间与转化率的数学模型进行系统仿真。并连接EscherichiacoliSFD4和PseudomonasdacunhaeDQp1的固定化细胞柱式反应器,试验了从石油裂解副产品反丁烯二酸连续生产L丙氨酸。
In the course of producing L-Alanine by enzyme, L-Alanine exerts a strong inhibition to the reaction. In this paper, the continuous L-Alanine production with packed-bed reactor is described. Pseudomonas dacunhae DQ-p1 is immobilized with k-Carrageenan, using the 1mol/L L-Aspartic acid as substrates, under 37 ℃ and pH 6.0. At the convertion rate of 95%, the efficiency of the immobilized cell and the reactor are 0.015g L-Alanine/g·immobilized cell·hr, 0.013g L-Alanine/ml·reactor·hr, respectively. Mathematic modeling and dynamic simulation of the system is implemented. We have also experimented to produce L-Alanine continuously from the fumaric acid (the by-roduct of petroleum decomposition)
