耐盐脱毛蛋白酶产生菌的研究──Ⅱ耐盐菌产酶条件的研究

Study on Salt-enduring Unhairing Proteases Producing Strain─Ⅱ The fermentation conditions of the salt-enduring strain

通过碳源、氮源实验以及碳氮配比实验研究，得到了耐盐菌适宜的产酶发酵培养基，培养基起始ｐＨ为７．０．接种菌龄１２小时种子液２％为宜。培养温度摄氏３０—３８度，培养时间４８—６０小时。该耐盐菌产酶可稳定在４１００－４４００ｕ／ｍｌ（培养液）。

The protease from No.813,a recent screened salt-enduring strain,is partially purified through a process of ammonium sulfate preciptation, dialysis,lyophilization.The optium pH and reaction temperature of the obtained proteases is 9.0 and 50℃ for casein hydrolysis,respectively the enzyme is stable over the pH range from 8 to 11,Ca2+ can enhance the stability against heat and pH of enzyme.Its activity was inhibited by Cr3+,Fe2+,Zn2+,Cu2+(1×10-3M),and stimulated by Mn2+.The enzyme is partially inhibited after treatment with EDTA(20mM) and PMSF(15mM).
