摘要
目的建立气相色谱法测定布格呋喃(AF-5)原料药的含量及有关物质。方法采用HP-5毛细管色谱柱(0.32 mm×30 m,0.25μm);柱温:程序升温,初始温度120℃,保持1 min,以10℃·min^-1升温至230℃,保持15min;进样器温度:200℃;检测器(FID)温度:250℃;载气:氮气;流速:1.0 mL·min^-1;分流比:20∶1。结果布格呋喃与有关物质的分离良好,方法的线性范围为0.01-1.0 g·L^-1(r=0.999 4),RSD〈2.0%(n=3),检测限为0.3ng。结论本法专属性强、简便、准确,易操作,适用于布格呋喃原料药的含量及有关物质的测定。
OBJECTIVE To develop a gas chromatography method for determining buagafuran(AF-5) and its related substances.METHODS The chromatographic analysis was performed using HP-5 capillary column(30 mm×0.32 mm,0.25 μm) with column temperature programmed from 120 ℃(holding for 1 min) to 230 ℃(holding for 15 min)by 10 ℃·min^-1.Injector temperature:200 ℃,Detector(FID)temperature:250 ℃,Carrier gas:nitrogen,Flow rate:1.0 mL·min^-1,Split ratio:20∶1.RESULTS Buagafuran and its related substances was separated.The linear range was 0.01-1.0 g·L6-1(r=0.999 4),RSD〈2.0%(n=3),the limit of detection was 1 ng.CONCLUSION The method is selective,simple,accurate and suitable for the determination of buagafuran and its related substances.
