摘要
目的在药物诱发抑郁模型上观察3,6′-二芥子酰基蔗糖(3,′6-d isinapoyl sucrose,DISS)的抗抑郁作用及可能的作用机制。方法采用小鼠5-羟色胺酸(5-hydroxytryptophan,5-HTP)增强实验,小鼠育亨宾(yohimb ine,YOH)毒性增强实验,小鼠阿朴吗啡(apomorph ine,APO)诱导体温下降实验探讨DISS的抗抑郁作用及可能的作用环节,用小鼠自主活动监测仪记录小鼠自发活动行为。结果在小鼠5-HTP增强实验模型上,给予DISS,对5-HTP诱导的小鼠甩头行为具有显著增强作用。在小鼠YOH毒性增强实验模型上,DISS可增强YOH毒性作用。在小鼠APO诱导体温下降实验模型上,ig给予DISS有显著性的拮抗体温降低的作用。结论DISS在药理学抑郁模型有显著的抗抑郁活性。并且其抗抑郁活性与增强5-羟色胺(5-HT)和去甲肾上腺素能(NE)神经功能均有关。DISS在抗抑郁有效剂量范围内无中枢兴奋或抑制性作用。
OBJECTIVE To explore the antidepressant effects and the possible mechanism of 3′,6-disinapoyl sucrose(DISS) in pharmacological depression models.METHODS The 5-hydroxytryptophan potentiation test,yohimbine toxicity potentiation test,apomophine induced hypothermia test in mice were used in this study.RESULTS DISS significantly increased the head-twitches symptom in the 5-hydroxytryptophan(5-HTP) potentiation test mice and the yohimbine toxicity in mice.Also,DISS showed significantly antagonized effect on the apomorphine(16 mg·kg-1,ip) induced hypothermia in mice.CONCLUSION DISS has notable antidepressant effect in pharmacological depression models,which is closely related to the potentiation of central 5-HT system and NE system.And antidepressant curative dose of DISS shows no exciting-or inhibiting-effect on central nervous system.
