合并应用抗病毒药物患者血浆中盐酸美沙酮的测定

The method of determination of methadone hydrochloride in plasma by HPLC in patients of anti-virus treatment

目的:建立测定应抗病毒药物患者血浆中盐酸美沙酮含量的方法。方法:采用C18ODS色谱柱(6.0mm×150mm,5μm),流动相为乙腈-磷酸二氢钾(38:62),检测波长为220nm。结果:方法回收率为94.82～101.64,日内RSD为2.04～2.36,日间RSD为4.11～6.76,线性范围为36～1152μg.L-1。结论:本法简便、准确,重复性好。

OBJECTIVE The HPLC method was established for determination of methadone hydroehloride in plasma in patients of anti-virus treatment. METHODS The HPLC method was developed under the following conditions：Column, C18 ODS （4. 6 mm × 150 mm, 5 μm）. The mobile phase was acetonitrile-monopotassium phosphate （38：62）. The wavelength of the UV detector was set at 220 nm. RESULTS The recovery was 94. 82% - 101.64% （n = 6）. The interday validation was 2. 04%- 2. 36% . The intraday validation was 4. 11%-6. 76%. The linear range was from36-1152μg·L^-1. CONCLUSION The method is simple, accurate and reproducible.
