4株PRRSV GP5蛋白编码序列的序列测定和特性分析 被引量:3

摘要 针对PRRSV ORF5基因组设计了1对特异性引物,利用RT-PCR的方法,对在南方地区分离的4株PRRSV ORF5基因进行了基因扩增,克隆到pMD18-T载体后测序,获得了4株PRRSV ORF5基因全长603bp的核苷酸序列。利用生物信息学软件对4株PRRSV ORF5基因序列进行了遗传进化分析,结果显示GD1、GD2、GD3、GD4株与美洲型参考株(VR-2332)的核苷酸序列同源性分别为87.1%、87.2%、87.1%和86.9%。利用系统进化树分析表明GD1、GD2、GD3、GD4株均属于美洲型。进一步采用序列分析软件对病毒结构蛋白推导糖基化位点比较,结果表明不同毒株GP5糖基化位点在基序和位置上均有所不同,这可能与毒株的毒力、免疫原性的差异相关,这些为更好地了解PRRSV的分子流行病学特征和抗原变异规律提供依据,同时为研制基因工程疫苗奠定了基础。
