卵巢上皮性肿瘤中基质金属蛋白酶MMP-9及其组织抑制剂TIMP-1 mRNA的表达及意义 被引量:2

摘要 目的:探讨基质金属蛋白酶(MMP-9)及其组织抑制物(TIMP-1)mRNA与卵巢上皮性癌的发生、发展及侵袭、转移的关系。方法:采用RT-PCR技术检测38例卵巢上皮性癌、15例卵巢交界性上皮性肿瘤、16例卵巢良性上皮性肿瘤及11例正常卵巢组织中MMP-9及TIMP-1 mRNA的表达,并分析其与各临床病理参数的关系。结果:MMP-9mRNA在卵巢上皮性癌及卵巢交界性上皮性肿瘤组织中的阳性表达率分别为76%、44%及表达水平(分别为154.14±4.42,96.87±3.04)均显著高于卵巢良性上皮性肿瘤及正常卵巢组织(分别为13%,9%和8.26±2.67,2.341.02;P均<0.05);且MMP-9 mRNA在晚期卵巢上皮性癌组织中的阳性表达率和表达水平明显高于早期(P<0.05)。TIMP-1 mRNA在卵巢上皮性癌及卵巢交界性上皮性肿瘤组织中的阳性表达率(分别为68%,38%)及表达水平(分别为126.35±3.81,90.62±5.54)均显著高于卵巢良性上皮性肿瘤及正常卵巢组织(分别为13%,9%和8.32±2.02,2.63±1.35;P均<0.05);TIMP-1 mRNA在卵巢上皮性癌组织中的阳性表达率和表达水平与各临床病理参数均无相关性(P>0.05)。结论:MMP-9 mRNA的高表达及MMP-9/TIMP-1平衡的调节失衡导致卵巢上皮性肿瘤的发生、发展;MMP-9 mRNA的高表达对卵巢上皮性癌的预后有意义。 Objective:To investigate the mRNA expression of matrix metalloproteinase - 9 (MMP- 9)and tissue inhibitor of matrix metalloproteinase - 1 (TIMP - 1 ) and their relations to clinicpathologic factors in epithelial ovarian tumor. Methods: RT - PCR was performed to assay the mRNA level of MMP - 9 and TIMP - 1 in 38 cases of epithelial ovarian cancer, 15 borderline epithelial ovarian tumor, 16 benign epithelial ovarian tumor and 11 normal ovaries. Results: The positive rate ( 76 % ,44%, respectively) and semi - quantitative level ( 154.14 ± 4.42,96.87 ± 3.04, respectively) of MMP- 9mRNA were significantly higher in epithelial ovarian cancer and borderline tumor compared with benign tumor and normal ovary ( 13 % ,9%, respectively and 8.26 ± 2.67,2.34 ± 1.02 respectively ; P 〈 0.05 ) and expression level of MMP - 9 mRNA in patients with stage Ⅲand Ⅳwas significantly higher than in stage Ⅰ and Ⅲ (P 〈 0.05 ). The positive rate (68 % ,38%, respectively) and semi - quantitative level ( 126.35 ± 3.81,90.62 ± 5.54,respectively) of TIMP- 1mRNA were significantly higher in epithelial ovarian cancer and borderline tumor compared with benign tumor and normal ovary ( 13% ,9% and 8.32 ±2.02,2.63 ± 1.35 ,respectively; P 〈0.05). The positive rate and semi - quantitative level of TIMP - 1mRNA had no correlation with clinicpathologic factors in epithelial ovarian tumor. Conclusion:The overexpression of MMP - 9 and the imbalance between MMP - 9 and TIMP - 1 showed significant relevance to the tumorigenesis,invasion and metastasis of epithelial ovarian tumor. MMP - 9 may have prognostic significance in epithelial ovarian cancer.
