摘要
目的:研究CD147和MMP-2在大肠癌组织中的表达和大肠癌患者血清中的水平变化,探讨其与大肠癌侵袭和转移的相关性,及二者间的相互联系。方法:应用免疫组织化学(SABC)法结合图像分析软件检测44例大肠癌、癌旁和正常对照组织中CD147和MMP-2的表达及半定量测定。应用酶联免疫吸附实验(ELISA)测定44例大肠癌患者血清中CD147和MMP-2的水平,并设36例健康对照。结果:大肠癌组织CD147和MMP-2的表达显著高于癌旁和正常对照组织,其表达与肿瘤分型、分化程度未见相关,而与转移密切相关。大肠癌患者血清CD147和MMP-2水平显著高于健康对照组,且伴转移组血清中CD147水平显著高于无转移组;低分化组中MMP-2水平显著高于高、中分化组。两者在组织定量和血清水平呈正相关。结论:大肠癌组织、血清中CD147高表达及高含量在一定程度上可以反映大肠癌发展程度及转移状态,其促MMP-2激活和表达增高的作用可能是其中一个重要因素。
Objective: To study the expression and serum levels of CD147 and MMP-2 protein in patients with colorectal cancer, to investigate the correlation between CD147 and MMP-2, and to explore the relationship of CD147 and MMP-2 with the invasion and metastasis of malignant tumor. Methods: Immunochemical staining was performed to detect the expression of CD147 and MMP-2 protein in colorectal cancer tissues, adjacent tissues, and normal controls. Semi-determination was performed by image analysis software. ELISA was used to detect CD147 and MMP-2 in the peripheral blood of patients (44 cases of colorectal cancer) and healthy controls (30 cases). Results: The expression of CD147 and MMP-2 in cancer tissues was significantly higher than in cancer tissues than in adjacent tissues and normal tissues, with no correlation with tumor types and differentiation. But the expression of CD147 and MMP-2 was much higher in patients with metastasis than in those without metastasis (P〈0.05). Serum levels of CD147 and MMP-2 were significantly higher in cancer patients than in the normal controls (P〈0.05). The serum levels of CD147 and MMP-2 were associated with tumor differentiation and Dukes' stage. The serum level of CD147 was positively correlated with MMP-2 in patients with colorectal cancer. Conclusion: The expression of CD147 is closely correlated with the malignant degree and metastasis in colorectal cancer, probably through activating and enhancing MMP-2 expression.
