摘要
目的由线栓法大脑中动脉闭塞致脑缺血叠加寒冷诱导建立血瘀证表征模型,通过与寒凝血瘀证表征模型和脑缺血诱导血瘀证表征模型比较,选择一种最合适的血瘀证表征模型。方法选择大脑中动脉闭塞(MCAO)大鼠模型、冰水冷冻大鼠模型、大脑中动脉闭塞叠冰水冷冻大鼠模型,进行脑缺血后1、3、5、7d不同时间点大鼠证候模型表征变化(舌、舌下脉络)及其对模型大鼠进行行为学、血液流变学检测。结果脑缺血后3dMCAO组、冰水组与冰水MCAO组大鼠的舌质色度值与正常组比较均存在统计学差异(P<0.05);脑缺血后5d冰水MCAO组大鼠的舌质色度值与正常组比较存在统计学差异(P<0.05);脑缺血后3d冰水MCAO组舌质色度值为各时间点最高值(舌质色度值60.12±5.15),但各组间舌质色度值比较统计学无差异(P>0.05)。各组血液流变学统计结果显示,MCAO组、冰水冷冻组、冰水MCAO组全血及还原黏度、血球压积、血浆黏度、聚集指数、变形指数均较正常组升高,部分时间点值升高与正常组比较有统计学差异(P<0.01,P<0.05),其中又以冰水MCAO组脑缺血后3d血液黏度、血球压积、血浆黏度、聚集指数均为各组最高值,变形指数为各组最低值。结论在3种模型中,大脑中动脉闭塞叠加寒冷模型可能为最合适的诱导血瘀证表征模型的动物模型。
Objective To establish an animal model with blood stasis syndrome by combination of cerebral ischemia and cool irritation, and compare it with two existing animal models with blood stasis syndrome in order to select an optimal exosymptom model of blood-stasis syndrome. Methods Three rat models of blood-stasis syndrome were selected, including middle cerebral artery occlusion model (MACO), cool irritation model (CI) and combination of middle cerebral artery occlusion and cool irritation model(MACO&CI). The exosymptoms of blood stasis syndrome were observed at different time points after cerebral ischemia (on 1^st, 3^rd, 5^th day and 7^th day ). Ethological and haemorheological detection were performed. Results Compared with the normal group, tongue color index values (TCIV) of rats were high 3days after cerebral ischemia in the MCAO, CI and MACO&CI groups, with significant differences(P 〈 0.05) ; only high in the MACO&CI group 5 days after cerebral ischemia ( P 〈 0.05 ).TCIV of the MACO&CI group was the highest (60.12±5. 154) 3 days after cerebral ischemia in all time points. The whole blood viscosity, reduced viscosity, plasma viscosity, hematocrit, index of erythrocyte aggregation and deformity index of RBC of rats in the MCAO, CI and MACO&CI groups were higher than those in the normal group, with significant differences at some time points (P 〈 0.05 ). Among all groups and all time points, in the MACO&CI group 3 days after cerebral ischemia the blood viscosity, hematocrit, plasma viscosity and index of erythrocyte aggregation were the highest and the deformity index of RBC was the lowest. Conclusions The animal model of combination of cerebral ischemia and cold stimulation was one rational exosyndrome model of blood stasis syndrome.
