摘要
以大黄素为标准品,建立了有效的薄层色谱(TLCS)测定大黄素在何首乌中的含量的方法。TLCS检测条件为:取硅胶G(60型),加入适量0.1%羧甲基纤维素钠(CMC-Na),制成厚0.5mm的薄层板,105℃活化30min,备用。展开剂:石油醚(30-60)-甲酸乙酯-甲酸(15∶5∶1)萃取(日光灯或紫光灯下检视),取上层液。展距67cm,λ366nm荧光灯下显黄色荧光。线性化系数X=3,狭缝1.2mm×1.2mm。该方法切实可行,大黄素具有良好的线性关系,测试结果比较准确,为大黄素含量测定奠定了基础,并可用来控制原药材何首乌及其制剂质量。
TLCS were used for the quantitative determination of emodin in Polygonum mutiflorum Thunb. 0.5mm lamella was made by adding 0.1% CMC-Na into silica gel G(60 type) ,activated at 105℃ for 30min on standby. Spray reagent: light petroleum (30-60)- ethyl formate- formic acid ( 15:5:1 ), extracte and get superstratum liquid. Expanding distance 6-7cm, appear yellow fluorescence at λ366nm fluorometry. Linearization coefficient X = 3, parochial aperture 1.2mm × 1.2mm. The result showed that this method was simple, accurate and reliable. The TLCS analysis technology could provide more complete and reliable evaluation for the qualities of Polygonum mutiflorum Thunb.
