薄层色谱法检测何首乌大黄素方法的建立

Analysis of emodin in Polygonum mutiflorum Thunb by TLCS

以大黄素为标准品,建立了有效的薄层色谱（TLCS）测定大黄素在何首乌中的含量的方法。TLCS检测条件为：取硅胶G（60型）,加入适量0.1%羧甲基纤维素钠（CMC-Na）,制成厚0.5mm的薄层板,105℃活化30min,备用。展开剂：石油醚（30-60）-甲酸乙酯-甲酸（15∶5∶1）萃取（日光灯或紫光灯下检视）,取上层液。展距67cm,λ366nm荧光灯下显黄色荧光。线性化系数X=3,狭缝1.2mm×1.2mm。该方法切实可行,大黄素具有良好的线性关系,测试结果比较准确,为大黄素含量测定奠定了基础,并可用来控制原药材何首乌及其制剂质量。

TLCS were used for the quantitative determination of emodin in Polygonum mutiflorum Thunb. 0.5mm lamella was made by adding 0.1% CMC-Na into silica gel G（60 type） ,activated at 105℃ for 30min on standby. Spray reagent： light petroleum （30-60）- ethyl formate- formic acid （ 15：5：1 ）, extracte and get superstratum liquid. Expanding distance 6-7cm, appear yellow fluorescence at λ366nm fluorometry. Linearization coefficient X = 3, parochial aperture 1.2mm × 1.2mm. The result showed that this method was simple, accurate and reliable. The TLCS analysis technology could provide more complete and reliable evaluation for the qualities of Polygonum mutiflorum Thunb.
