胡萝卜的愈伤组织诱导及其几种酶和可溶性蛋白质的变化

摘要 胡萝卜肉质根韧皮部培养于附加不同激素的MS培养基上均可产生愈伤组织,但以附加QBS培养基上愈伤组织诱导率低,且愈伤组织在第24天后逐渐褐化。随着脱分化的过程,外植体及其愈伤组织中的多酚氧化酶、抗坏血酸氧化酶和过氧化物酶的活性以及可溶性蛋白质的含量都逐渐升高。附加QBS的培养基上所生长的愈伤组织中,这三种酶的活性以及多酚氧化酶对65℃热的稳定性都明显低于附加NAA、6-BA的培养基上的。 Cultured on MS medium with one of some hormones, carnose root explants of Daucus carota can produce calluses, The induction ratio is lower than usual when they are cultured on MS medium with QBS, and the induced callus turn tawny gradually after 24 days of culturing. In the process of dedifferentiation. the activity of Polyphenol oxidase, Ascorbic acid oxidase, and Peroxidase increases in the explants and calluses and so does the content of soluble protein. The activity of the three enzymes is lower in the calluses on MS medium with QBS than in those on MS medium with NAA or 6-BA. The resistance of Polyphenol oxidase to 65℃ heat is weaker when cultured on MS medium with QBS than it is on MS medium with NAA or 6-BA.
