摘要 目的探讨甲状腺过氧化物酶(TPO)mRNA和肝细胞生长因子及其受体蛋白(HGF/c-Met)在良恶性甲状腺结节中的表达及意义。方法应用逆转录聚合酶链反应技术检测55例甲状腺结节新鲜标本的TPO mRNA丰度,采用免疫组织化学法检测600例甲状腺结节石蜡标本HGF/c-Met的表达。结果甲状腺癌HGF/c-Met表达率显著高于甲状腺良性结节,乳头状癌高于滤泡型癌(P<0.01)。甲状腺恶性结节TPOmRNA的阳性表达率显著低于甲状腺良性病变。结论HGF/c-Met和TPO的联合检测在良恶性甲状腺病变的鉴别中有一定应用价值。 Objective To explore the expressions and significance of the thyroperoxidase (TPO) mRNA, hepatocyte growth factor (HGF) and its receptor (c-Met) protein in thyroid diseases and implication. Methods Abundance of TPO mRNA was detected by reverse transcription-polymerase chain reaction technique in the samples from 55 thyroid fresh masses. HGF/c-Met expression was assayed with immunohistochemical method in 600 thyroid paraffin samples. Results The positive rates of HGF/c-Met protein expressions in thyroid carcinomas were significantly higher than those of benign thyroid diseases and the expressions were significantly between papillary and follicular thyroid malignancies (P 〈 0.01 ). The positive rates of TPO mRNA expression in thyroid carcinomas was significantly lower than those of benign thyroid diseases( P 〈 0.05). Conclusions The federate inspection of the TPO mRNA and HGF/c-Met in thyroid diseases can be valuable in diagnosing malignant and benign thyroid diseases.
