摘要
采用PAGE及SDS-PAGE法,分析了绵阳本地半夏属植物圆叶半夏的5个居群、狭叶半夏5个居群和中间过渡型半夏2个居群及虎掌等几个品种,以及采自湖北、平武半夏样品的POD同工酶、可溶性蛋白谱带及不同样品的POD活性;在此基础上通过对块茎可溶性蛋白谱带的UPGMA聚类分析,对15个样品的亲缘关系进行了初步探讨。
The difference of POD and soluble protein of Pinella, including 5 special populations of P.ternata,5 special populations of P.angustata, 2 transitions of P.ternata and P.angustata, P.pedatisecta from Mianyang, and Pinella from Hubei and Pingwu were analyzed by PAGE and SDS-PAGE. The activity of POD and molecular weight of soluble protein were also analyzed. Results showed that the patterns of POD and soluble protein in the tubers were more various than those in the leaves. Each species had their own features, and could be used as bio-chemical indexes for identifying these plants and their origins. Cluster analysis of soluble protein in the tubers, applying the method of UPGMA, showed that the 5 special populations of P.ternata and P.angustata, and the 2 special populations of the transitions of the two had the lowest genetic distance. The transitions of the two were nearer to P.angustata, and could be classified into 1 grope at level of GD 0.33. Pinella from Hubei and Pingwu had the higher genetic distance, which indicated that Pinella from the two places were a new species; P.pedatisecta had the highest genetic distance and classified into a single grope.
