摘要
目的探讨硒、维生素E(VE)和维生素C(VC)对经X射线照射后的细胞抗氧化损伤作用。方法利用碱性单细胞凝胶电泳法(彗星实验),观察面积和尾长2个指标,并进行分组设计,分析3种抗氧化剂硒、VE和VC不同水平的抗氧化作用以及二者或三者之间的交互作用。结果硒、VE和VC单因素作用下,不同水平的彗星平均面积和尾长间差异均有统计学意义(P<0.0001),硒与VC的交互作用差异有统计学意义(P<0.05)。结论硒、VE和VC分别可以拮抗因射线而产生的DNA损伤作用,其作用随浓度的增加而加强;硒与VC之间存在拮抗DNA损伤的协同交互作用。
Objective To explore the anti-oxidization effects of sodium selenite, vitamin E and C after X-ray radiation. Methods Different dosages of sodium selenite, vitamin E and C were added into cell culture after irradiation to observe changes in DAN damage and synergistic effects between the both or three antioxidants by means of comet assay. Results Sodium selentie, vitamin E and C can decrease cell DNA damage( P 〈 0. 001 ). Synergistic effect existed between ,sodium se- lenite and vitamin C(P 〈 0.05). Conclusion Sodium selenite, vitamin E, vitamin C can antagonize radiation DNA damage effectively, and the effect increases with the dose of the antioxidant. Synergistic effect exists between sodium selenite and vitamin C in antagonizing radiation DNA damage.
