人胚胎生殖腺嵴和肠背系膜来源的成纤维样细胞作为饲养层体外培养人胚胎生殖细胞方法的建立

摘要 目的:研究以来源于人胚胎生殖腺嵴和肠背系膜的成纤维样细胞为饲养层,体外培养人胚胎生殖细胞(hEG)。方法:分离培养孕5~9周人胚胎的生殖腺嵴和肠背系膜中的成纤维样细胞,以该细胞作为饲养层,体外原代培养来源于孕5~9周人胚胎的生殖腺嵴和肠背系膜组织的hEG。免疫荧光染色法、碱性磷酸酶染色法和体外分化实验用于鉴定所培养的hEG。结果:成功分离培养人胚胎生殖腺嵴和肠背系膜来源的成纤维样细胞,可传至25代以上,并保持状态稳定,3~15代人胚胎成纤维样细胞用于制备饲养层。21例hEG原代培养中,有7例获得明显增殖的hEG细胞集落,目前传到第10代,仍然保持碱性磷酸酶、胚胎阶段性表面抗原(SSEA)-1、SSEA-4和POU转录因子Octamer-4(Oct-4)的表达为强阳性,并具有向3个胚层细胞分化的能力。结论:成功利用人胚胎生殖腺嵴和肠背系膜的成纤维样细胞作为饲养层细胞,获得了能够增殖并保持未分化状态和多向分化潜能的hEG,建立了一种新的hEG培养方法,为hEG的体外培养建系奠定基础。
