摘要
目的利用蛋白芯片技术对鼠疫血清进行检测,分析其血清抗体谱。方法用制备好的蛋白芯片和未知的血清进行杂交后荧光检测并数据分析。结果发现具有抗原性,免疫原性的相关蛋白。结论通过分析血清抗体谱而得到新的具有免疫原性的蛋白作为潜在的候选疫苗组;得到新的可以用于鼠疫菌诊断的抗原,作为目前血清学检测的辅助诊断抗原;通过对系列血清抗体变化趋势以及不同种属间抗体谱的区别的分析,加深对鼠疫菌致病机制的认识。
Objective Using the technique of protein chip to exam the plague germs and analyze the plague germ serum antibody profile. Methods The prepared protein chip was detected by fluorescence and the data were analyzed after hybridized with unknown plague germ serum. Results GAP - associated protein was found with anfigenicity and immunogenicity. Conclusion According to the analysis of the serum antibody profile we can get the immunogenic protein which can be used as the candidate vaccine group, this is the first profits. Secondly we can also get the antigen which can assist the diagnosis of plague germ infection. Finally through the analysis of the changing tendency of the series serum antibodies and the differences among the antibody profile of different species,we can deepen our knowledge about plague germ pathogenesis mechanism.
