刺五加注射液对衰老模型大鼠学习记忆障碍及海马单胺类神经递质的影响 被引量:5

Effects of Acanthopanax Senticosus Injection on Learning and Memory Impairment and Monoamine Neurotransmitter of Hippocampus in Aging Model Rats

摘要 目的探讨刺五加注射液(Acanthopanax Senticosus injection,ASI)对喹啉酸(quinolinic acid,QA)致衰老模型大鼠学习记忆力及脑海马匀浆中单胺类神经递质(ACh、NE、5-HT)的影响。方法将60只大鼠随机分成对照组、假手术组、模型组、治疗组(ASI小剂量组、ASI中剂量组和ASI大剂量组)。除假手术组外,其他各组均以双侧海马CAI区注射喹啉酸法复制老年性痴呆模型大鼠。对照组用石杉碱甲(Huperzine A,Hup-A)治疗,治疗组用不同剂量ASI(50、100、200 mg/kg)治疗30 d后,以Morris水迷宫法测试学习记忆能力及空间探索能力,并测定海马匀浆中ACh、NE、5-HT的含量。结果ASI能显著缩短记忆缺失模型鼠寻找平台的潜伏期,减少错误次数;各治疗组海马匀浆ACh、5-HT、NE的含量均显著高于模型组而低于假手术组(P<0.05或P<0.01),且呈现剂量递增趋势。结论ASI能够改善衰老模型大鼠的记忆能力,其机理可能与动员海马单胺类神经递质有关。 Objective To study the effects of Acanthopanax Senticosus Injection(ASI) on learning and memory impairment and monoamine neurotransmitter(ACh,NE and 5-HT) of hippocampus homogenate in aging model rats.Metholds Total of 60 male Wister rats were randomized into 6 groups: control group,sham operation group,model group and treatment group,the latter was randomized into low,middle and high dose groups.All group anesthetized by chloral hydrate.After stereotaxis,the ageing models were build by injecting QA(dissolved into PBS) 2 μL into CAI district of double sides of hippocampus in those control group,model group and treatment group,the sham operation group were injected the same volume of PBS.One week after operation,NS,ASI or huperzine A was given(ig) to rats for 30d.The sham operation and model groups were given NS(2 mL).The treatment groups were given ASI(50,100,200 mg/kg).The control group were given Huperzine-A(0.3 mg/kg).The ability was tested by Morris water maze on learning,memory and space exploration in rats.ACh,NE and 5-HT were measured in hippocampus homogenate.Results The incubation period of seeking platform was decurtated and the errors was reduced in aging model rats by ASI;The treatment groups of ACh,NE and 5-HT of hippocampus homogenate were higher than model group,but lower than sham operation group(P〈0.05 or P〈0.01),strengthening effects with increasing dosage.Conclusion ASI can obviously improve learning and memory,the mechanism of action mechanism correlates to the mobilization of the monoamine neurotransmitter(ACh,NE and 5-HT) in hippocampus.
