摘要
目的探索黄疸大鼠脾脏Th1/Th2细胞因子、蛋白激酶C(PKC)αmRNA的表达情况及茵陈蒿汤的干预效应。方法大鼠随机分为3组:正常对照组、黄疸模型组、茵陈蒿汤组。ELISA法、原位杂交法检测大鼠脾细胞分泌IL-4、IFN-Υ水平及PKCαmRNA表达。结果黄疸模型组IFN-Υ/IL-4比值明显低于正常对照组(P<0.01),而茵陈蒿汤IFN-Υ/IL-4比值与正常对照组比较无显著差异(P>0.05)。黄疸模型组脾细胞PKCαmRNA表达明显高于正常组(P<0.01),而茵陈蒿汤治疗后,脾细胞PKCαmRNA表达明显减少。结论黄疸大鼠脾脏ThI/Th2细胞因子分泌呈紊乱状态,与PKCαmRNA表达水平有关,而茵陈蒿汤具有调节作用,可能是其治疗黄疸的机制之一。
Objective To investigate the expression of Th1/Th2 cytokines and PKCαmRNA of spleen cells and the effects of YCHT in jaundice rats.Methods The rats were divided randomly into 3 groups: the normal control group,jaundice model control group,YCHT group.ELISA assay and in site hybridization technique were used to detect the expression of IL-4 and IFN-Υ and PCKαmRNA respectively in spleen cells of rats.Results The specific value of IFN-Υ/IL-4 in jaundice model group was lower significantly than that in formal control group(P〈0.01),but there were indifference significantly between YCHT group and formal control group(P〉0.05).The expression of PKCαmRNA of spleen cells in jaundice model group was lower than that in formal control group,but the expression of PKCαmRNA of spleen cells decreased significantly by treating with YCHT.Conclusion The expression of Th1/Th2 cytokines of spleen cells show disorder in jaundice rat,it related to the expression of PKCαmRNA,but YCHT can regulat it,and it is one of the mechanismes on treating the YING jaundice.
