摘要 文章以马铃薯品种东农303的脱毒微型薯为外植体,利用农杆菌介导法将Bt-CryV基因转入马铃薯中,同时优化影响遗传转化的因素,包括激素种类与浓度、休眠期、脱菌抗生素种类与浓度、共培养时间、筛选方式等。获得21株Kan抗性植株进行PCR和PCR-Southern检测,其中10株为阳性植株,证明外源基因已经整合到植物的基因组上。 Bt-CryV gene was transferred into the potato cuhivar Dongnong 303 by agrobacterium-mediated method. Microtuber discs were used as explants. A regeneration system for potato transformation was established when several factors, such as the kind and concentration of different phytohormones and antibiotics, the dormant stages of microtuber, the time of co-culture, and the method of selection, were optimized. The 21 transgenic plants were regenerated and selected on MS basal medium supplemented with Kan 100 mg ·L^-1, of which 10 transformants were confirmed by the test of PCR and PCR-Southern Blot.
