痴呆模型大鼠背海马结构内淀粉样蛋白沉积的免疫细胞化学研究 被引量:27

摘要 用AICL3灌胃,建立大鼠老年性痴呆模型。用免疫细胞化学ABC法,观察了β-淀粉样蛋白(β-AP)样免疫反应(APLI)神经元在大鼠双侧背海马结构的分布、形态、大小和数量。结果显示在痴呆模型大鼠背海马结构各部均可见较多的APLI神经元。阳性反应产物主要位于神经无核周质和突起。细胞大小不等,多数APLI神经元胞体为梭形和锥形。背海马结构内APLI神经元主要分布在CA1、CA2、CA3和CA4区的锥体细胞层及齿状回多形细胞层,其中在齿状回和CA1区APLI神经元数量最多。说明海马齿状回、CA1区神经元结构的改变与老年性痴呆的发生有密切关系;同时也表明铝可引起β-AP在大鼠背海马结构神经元的沉积。 After pretreatment with aluminium chloride (P. O), the model of dementia was estab-lished. With immunocytochemical method of ABC,the expression of β-amyloid protein-like immunore-activity(APLI) in the dorsal hippocampal formation of rat was investigated. The results showed that large number of APLI neurons were found in bilat-eral dorsal hippocampal formation. The reaction products were 16cated in cytoplasm and processes.The size of APLI neurons is different. Most of the APLI cells were found to be of fusiform or pyrami-dal shape. The APLI neurons were distributed chiefly in pyramidal cell layer of CAl, CA2, CA3and CA4 region of dorsal.hippocampal formation and polymorphical cell layer of dentate gyrus: the APLI neurons were the most in dentate gyrus and CAl re-gion. These results suggest that the change in neu-ronal structure of dentate gyrus and CAl region is related to Alzheimer's disease, and that aluminium can cause AAP to accumulate in neurons of dorsal hippocampal formation in rat.
