TNF-α与PGC-1α影响成熟脂肪细胞分泌RBP4的研究

摘要 目的观察过氧化物酶增殖体激活受体辅激动子1α(PGC-1α)、肿瘤坏死因子-α(TNF-α)及两者联合对体外培养的成熟3T3-L1脂肪细胞分泌视黄醇结合蛋白4(RBP4)的影响。方法体外培养3T3-L1细胞,待细胞完全融合后两天,诱导分化为成熟脂肪细胞,使用重组PGC-1α腺病毒,对照病毒,以及重组TNF-α(10ng/ml),TNF-α联合PGC-1α腺病毒干预,采用放射免疫分析法检测24小时后成熟脂肪细胞RBP4的分泌量。结果①各干预组均能导致成熟脂肪细胞RBP4分泌的降低。②对照病毒组和PGC-1α腺病毒组引起RBP4的降低,两组间差别无统计学意义。③TNF-α组和TNF-α联合PGC-1α腺病毒组两组减少脂肪细胞分泌RBP4,差别无统计学意义。④两含有TNF-α组和两病毒组对RBP4的影响有统计学差异。结论PGC-1α对成熟脂肪细胞分泌RBP4无影响,TNF-α能抑制成熟脂肪细胞RBP4的分泌,两者对RBP4分泌的影响无相互作用。 Objective To explore the effects of tumor necrosis factor-or (TNF- α), Peroxisome proliferator- activated receptor- gamma coactivator - 1 α ( PGC - 1 α) and PGC - 1 α mixed with TNF - α on retinol binding protein 4 (RBP4) secreted by mature 3 T3 - L1 adipocytes in - vitro. Methods 3T3 - L1 preadipocytes were cultured and differentiated into the mature adipocytes after 2 days of confluenence. Recombinant PGC - 1α adenovirus, control virus, recombinant TNF - α(10ng/ml) , PGC -1α adenovirus mixed with TNF - α(10ng/ml) were added to the cuhure medium of fully differentiated adipocytes. Total protein of RBP4 secreted by these adipocytes was evaluated by radioimmunoassay (RIA) for 24h after. Results 1. Treatment of mature 3T3 - L1 adipocytes with all these different interventions resulted in the decreasion of RBP4 secretion. 2. Both of the control virus and recombinant PGC -1α adenovirus groups had an inhibitive effect on RBP4, but there's no statistical difference in the two groups. 3. There's no differences between the groups of TNF - α intervention and PGC - 1 ct mixed with TNF - α on the effects of reduction of RBP4 secretion. 4. There's statistical difference in the two groups between TNF -α and virus intervention groups. Conclusions PGC - 1α has no effect on RBP4 secreted by mature 3T3 -L1 adipocytes while TNF -α can inhibite the protein from adipocytes , and they have no interaction with each other.
