果子狸细小病毒分离及VP2基因的进化分析 被引量:1

摘要 采用F81细胞从患有肠炎的果子狸粪便样品中分离出2株病毒,经形态学、血清学、生物学和病毒分子生物学鉴定分离的病毒为CPV突变株。对2株细小病毒VP2基因进行克隆和基因进化分析表明,2株病毒与肉食动物细小病毒有很高的同源性,属于CPV-2a突变株,分别命名为PV/PL/HeN02/08和PV/PL/HeN03/08。进一步分析表明,PV/PL/HeN03/08株接近于CPV-2a型毒株,仅在第297位氨基酸发生A→S突变;PV/PL/HeN02/08株在第300位氨基酸发生G→A突变,第305位氨基酸发生Y→D突变,除第87位氨基酸外,该毒株更接近于CPV-2型毒株。 Two parvovirus strains were isolated from feces of civets using F81 cells. Both of them were designated as mutant strains of CPV, through a series of identification assays, such as morphological, serological, biological and molecular virology assays. These strains were named as PV/PL/HeN02/08 and PV/PL/HeN03/08 respectively. Sequence analysis of the VP2 genes showed that high homology to other carnivore parvovirus strains. Comparison of the predicted amino acid sequences of VP2 genes suggested that PV/PL/HeN02/08 strain was corresponded to CPV type 2a and possessed an mutation at position 297 (A→S), while the PV/PL/HeN03/08 strain was corresponded to CPV type 2 and contained the mutations at positions 300 (G→A) and 305 (Y→D) respectively.
