摘要 目的:研究AB-8大孔树脂分离纯化桑叶黄酮的工艺条件。方法:通过静态吸附-解吸实验筛选最佳大孔树脂,采用L9(34)正交试验法对AB-8大孔树脂分离纯化桑叶黄酮的因素进行考察,以总黄酮含量为考核指标,确定最佳工艺条件。结果:较优工艺条件为:采用AB-8型大孔树脂,取桑叶黄酮提取液(含生药0.5g/mL)上样,树脂用量为3倍生药量,洗脱剂采用50%乙醇,用量为16倍生药量。结论:该工艺条件科学合理,可有效地用于桑叶黄酮的分离富集。 Objective: To study technique condition for separating total flavonoids from mulberry by AB - 8 macroporus resin. Methods: With the yield of total flavonoid as a maker, the static and dynamic adsorptiort/desorption methods and orthogonal design L9 (3^4) were used to investigate different factors such as concentration of alchol, ratio of alchol to raw material and ratio of resin to raw material so as to get the optimum process. The content of total flavonoid is determined by UV - Vis sepectrophotometer. Results : In the macroporus resin seperation process, ratio of resin to raw material was the primary factor, and the optimum condition was that the extrated solution (containing 0.5g/mL raw material) was passed through AB -8 resin( 3 times the weight of raw material) chromatographic column, after which, eluted it with distilled water to be almost colorless, then eluted it with 16 -fold of 50% alchol. Conclusion: The technique condition is seentific and reasonable. It can be used to separate and purificate total flavonoids from mulberry.
