摘要
为探讨人重组γ-干扰素(hrlFN-γ)能否抑制抗原引起嗜酸性粒细胞(EOS)在支气管哮喘患者气道的聚集。我们选择10例过敏性哮喘患者以纤支镜直接将hrlFN-γ及过敏原注入左舌叶或右中叶的肺段支气管,将生理盐水及过敏原注入对侧肺相应的肺段作为对照。24h后复以纤支镜取BALF作细胞计数及分类,并以免疫组化法测定IL-5和GM-CSF阳性细胞的百分率。结果hrIFN-γ组BALF中EOS数明显低于对照组,且IL-5和GM-CSF水平也都明显降低。结论hrIFN-γ由于能抑制了IL-5和GM-CSF的产生从而抑制了EOS对哮喘患者气道的浸润。
This research was to investigate whether human recombinant gamma interferon (hrIFN γ) was capable of inhibiting antigen induced airway eosinophil infiltration in patients with allergic bronchial asthma 10 atopic asthmatic subjects underwent an initial bronchoscopy during which hrIFN-γ and dust mite were instilled into a subsegment of the left lingula or right middle lobe At the same time,acting as controls,normal saline solution and dust mite were instilled into a subsegment of the contrlateral lung (middle lobe or lingula) A second bronchoscopy was performed 24 hours later, and bronchoalveolar lavage fluids (BALF) were respectively obtained from the above mentioned treated sites for BALF cell count and cell classification,and the detection of the percentages of IL 5 positive cells and GM CSF positive cells.Results:The number of eosinophils in BALF obtained from hrIFN γ treated sites (11 6±3 2)×10 6 was significantly lower than that (35 9±8 2)×10 6 of the control sites (P<0 01).Also,the percentages of both IL 5 positive cells and GM CSF positive cells obtained from hrIFN γ treated BALF were lower than those of the control sites(P<0 01) Conclusion:Local administration of hrIFN γ can prevent antigen induced eosinophil infiltration of the asthmatic airways by inhibiting the production of IL 5 and GM CSF
