鱼藤酮对小鼠脑神经递质多巴胺及其代谢产物的影响

摘要 研究了注射鱼藤酮对C57BL/6N小鼠神经递质多巴胺的影响。给小鼠皮下注射鱼藤酮(10mg/kg)两次,间隔2h,在第2次注射8h后分离脑组织样品,并应用高效液相色谱仪配电化学检测器,测定了小鼠脑内多巴胺(DA)及其主要代谢物3,4-二羟苯酰乙酸(DOPAC)和高香草酸(HVA)的水平。结果表明,第2次注射8h后,鱼藤酮处理组的脑内DA、DOPAC和HVA水平分别比对照组高40.5%、544.4%和168.2%,差异显著(P<0.01)。实验显示,鱼藤酮对多巴胺能神经系统具有毒性作用。 The effect of injection rotenone on the brain levels of dopamine (DA) and its metabolites (DOPAC and HVA) in C57BL/6N mice was studied. The mice were given two subcutaneous injections of rotenone (10 mg/kg) at 2 h interval between injections,and were killed at 8 hr after the second injection. The brain tissue was weighed and placed in a 5 mL tube containing ice-cooled perchloric acid (2 mL, 0.4 mol/L), homogenized for 1 rain at 20 000 r/min using an Ultra Turrax and centrifuged at 16 000 g for 12 min at 4℃. The supernatant was passed through a 0.2 μm filter and either directly analyzed or frozen at - 70℃ until analysis. The DA and two of its metabolites were determined by HPLC with ECD. Results showed the percentage of increasing reached 40.5 % ( DA), 544.4% (DOPAC),and 168.2% (HVA) as compare to the control (P 〈0.01) after the last injection. The rotenone has been suggested a potential nueurotoxicant that induced dopaminergic neurons to release dopamine and its metabolites in the mice.
