青石棉诱发的转化细胞p53基因突变分析

摘要 为探讨青石棉与p53基因突变之间的关系,采用PCR-SSCP分析方法,对8株青石棉诱发的BALB/c3T3转化细胞进行p53基因的突变检测和分析。结果:从8株转化细胞p53基因上的11个外显子中共发现7处突变,其中2个位于外显子4-6,5个位于外显子9-11;大多数检出的异常表现为比野生型多1条带;突变随机地分布于各个青石棉剂量组。由此表明,p53基因突变与体外转化实验中所用的青石棉剂量无关,检出的突变多位于外显子9-11内,与人类肿瘤中常见p53基因突变多发生在外显子5-8的情形有所不同。 This study sought to address the relationship between crocidolite and p53 gene mutation.The mutations of p53 gene in 8 BALB/c 3T3 cell lines transformed by crocidolite were analysed.Altogether 11 exons of the gene from 8 cell lines were detected by PCR SSCP. 7 alterations were found;2 of them were located in exon 46,and 5 in 911.Most of the mutations(5/7) were of one more band than that of wild cell from SSCP,and alterations were randomly scattered among the crocidolite doses groups.The results suggest that the presence of a p53 alteration is not related to the dose of crocidolite used.Besides,p53 mutation may occur in a relatively later period of the growth of the transformed cell lines.The results also showed that the mutations occurred predominantly in exons 911.This was different from that seen in human mesothelioma where mutations in the exon 58 of p53 gene were more frequently observed.
