摘要
目的:研究姜黄素对日本血吸虫病小鼠肝纤维化的影响。方法:建立血吸虫病肝纤维化小鼠动物模型,随机分为4组:正常对照组、感染对照组、吡喹酮治疗组、姜黄素加吡喹酮治疗组。应用HE染色、免疫组化法及多媒体病理图文定量分析,观察各实验组小鼠肝脏的病理改变及I、Ⅲ型胶原表达的变化,并测定血清谷丙氨酸转移酶(ALT)、谷草氨酸转移酶(AST)、透明质酸(HA)及层黏连蛋白(LN)的水平以反映肝细胞损伤及肝纤维化的程度。结果:姜黄素加吡喹酮治疗组小鼠肝脏纤维组织的增生明显减少,I、Ⅲ型胶原的含量及血清ALT、AST、HA和LN的水平明显低于血吸虫感染对照组及吡喹酮治疗组。结论:姜黄素能减轻血吸虫病时肝细胞的损伤并有抗肝纤维化作用。
OBJECTIVE To study the effect of curcumin on liver fibrosis of mice caused by Schistosoma japonicum infection. METHODS A total of 40 mice were randomly and averagely divided into group A, B, C and D. The mice in group A served as normal controls, while those in the other three groups were infected with schistosoma japonicum to induce the model of liver fibrosis. The mice in group C and D were treated with praziquantel, and praziquantel plus curcumin, respectively. Histological specimens were stained with HE and studied microscopically to observe the pathologic changes of liver fibrosis. Expression of type Ⅰ and type Ⅲ collagen was detected by immunohistochemical technique and multimedia color pathographic analysis system. The levels of serum ALT,AST,HA and LN were compared after being treated. RESULTS Fibrous tissue accrementition,the content of type Ⅰ and type Ⅲ collagen reduced significantly in curcumin plus praziquantel group compared to infected group and praziquantel group. The level of serum ALT, AST, HA and LN improved obviously in curcumin plus praziquantel group compared to control group and praziquantel group. CONCLUSION curcumin can relieve the liver fibrosis caused by schistosome.
