组织蛋白酶B和趋化因子受体4在乳头状甲状腺癌的表达及在侵袭转移中的作用 被引量:4

The expression of cathepsin B and CXC chemokine receptor 4 in papillary thyroid carcinoma and their contribution to invasion and metastasis of papillary thyroid carcinoma

摘要 目的观察组织蛋白酶B(CB)、趋化因子受体4(CXCR4)mRNA和蛋白在乳头状甲状腺癌(PTC)中的表达,探讨其在乳头状甲状腺癌侵袭和转移中的作用。方法采用逆转录一聚合酶链反应(RT-PCR)及免疫组织化学SP法检40例PTC,10例正常甲状腺中CB、CXCR4mRNA和蛋白的表达。结果CB、CXCR4mRNA和蛋白在正常甲状腺中均为阴性表达;CB、CXCR4 mRNA在PTC中阳性表达率82.5%(33/40)、80%(32/40);CB、CXCR4蛋白在PTC中阳性表达率65%(26/40)、60%(24/40);CB,CXCR4mRNA表达与蛋白表达之间呈显著正相关(r=0.352,P〈0.05;r=0.357,P〈0.05),且均与侵润深度,淋巴结转移密切相关(P〈0.05);CB,CXCR4蛋白表达呈正相关(r=0.471,P〈0.05)。结论CB、CXCR4在乳头状甲状腺癌的侵袭和转移中发挥促进作用。 Objective To observe the expression of cathepsin B ( CB ) and CXC chemokine receptor 4 (CXCR4) mRNA and protein in papillary thyroid carcinoma (PTC) and explore their contribution to invasion and metastasis of papillary thyroid carcinoma. Methods The expression of CB and CX- CR4 mRNA and protein in 40 cases of PTC and 10 cases of normal thyroid was detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemical SP method. Results The expression of CB and CXCR4 mRNA and protein was negative in normal thyroid ; The positive expression rate of CB and CXCR4 mRNA in PTC was 82.5% (33/40) and 80% (32/40) respectively;The positive ex- pression rate of CB and CXCR4 protein in PTC was 65% (26/40) and 60% (24/40) respectively. There was a positive correlation between CB and CXCR4 mRNA and protein ( r = O. 352, P 〈 0.05 ; r = 0. 357, P 〈 0.05 ). The expression of CB and CXCR4 mRNA and protein was significantly related to depth of invasion and lymph node metastasis ( P 〈 0.05 ). There was a positive correlation between the expression of CB and CXCR4 protein (r = 0.471 ,P 〈 0.05 ). Conclusion CB and CXCR4 play a positive role in invasion and metastasis of papillary thyroid carcinoma.
