高效液相色谱法测定蜂胶液中的6种有机酸 被引量:10

摘要 建立了蜂胶中6种有机酸的高效液相色谱测定方法.固定相为Eclipse XDB-C18柱(4.6mm×150mm;5μm),流动相为乙腈-0.4%的冰乙酸溶液(φ=30∶70,pH=2.93),流速0.8mL/min,柱温28℃.检测波长采用设定波长程序的方法,样品中6种组分的加标回收率在89.35%~104.5%之间,绿原酸、咖啡酸、香草酸、阿魏酸、苯甲酸、肉桂酸的线性范围分别为0.022~44,0.025~50,0.029~58,0.022~44,0.0414~62.1,0.0444~66.6μg/mL,检测限分别为4.55,3.95,8.7,6.29,10.35,3.92ng/mL,相对标准偏差小于3%.该法简便、快速、灵敏,可用于蜂胶中6种有机酸的同时分离测定. A reversed-phase high performance liquid chromatographic method was developed for the determination of six organic acids in propolis liquid. The six compounds were analyzed by an Eclipse XDB-C18 column (4. 6mm× 150 mm;5 m), with a mobile phase of acetonitrile -0. 4 acetic acid glacial (φ= 30:70, pH=2. 93), a flow rate of 0. 8 mL/min and a column temperature of 28℃. The detection wavelength was performed according to time program. 0-2.6 min,325 nm;2.6-3. 0 min,260 nm;3.0-4. 0 min,325 nm; and 4.0--9.0 min,280 nm. The recoveries of sample added standard ranged from 89.35% to 104.5%, and the relative standard deviation was below 3%. The method proved to be simple, rapid and highly sensitive according with the results obtained by HPLC-UV detection and is recommended for the simultaneous separation and determination of the above-mentioned six organic acids in propolis liquid.
