摘要
目的探讨环氧合酶2(COX-2)基因多态性与膀胱癌易感性的关系。方法采用病例对照研究方法,以聚合酶链反应限制性片段长度多态性技术(PCR—RFLP)及错配聚合酶链反应-限制性片段长度多态性技术(PIRA-PCR)分别检测180例膀胱癌患者和180例非肿瘤患者COX-2基因-765G/C、-1195G/A和-8473T/C3个多态位点,采用X2检验比较不同基因型与膀胱癌风险的关系。结果膀胱癌组和对照组COX-2—765G/C及1195G/A2个位点的3种基因型频率分布差异无统计学意义(P=0.582,P=0.270);2组间COX-2—8473T/C位点的3种基因型分布差异有统计学意义(P=0.041),携带突变等位基因C的个体(-8473TC+CC)患膀胱癌的风险显著降低(OR=0.56,95%CI=0.35~0.88)。结论COX-2—765G/C以及-1195G/A可能与膀胱癌易感性无关,COX-2—8473T/C与膀胱癌易感性相关。
Objective To study the relationship between cyclooxygenase 2 (COX-2) polymorphisrns and the susceptibility of bladder cancer. Methods Polyrnerase chain reaction-restricted fragments length polyrnorphisrn (PCR-RFLP) and the primer-introduced restriction analysis (PIRA-PCR) assay were used to genotype the COX-2 -765G/C, 1195G/A and -8473T/C polyrnorphisrns in a casecontrol study of 180 bladder cancer cases and 180 cance-free controls in a Chinese population. Results The distribution of the genotype frequencies of -765G/C and -1195G/A were not statistically different between the cases and controls (P:0. 582 for -765G/C and P=0. 270 for -1195G/A). Polymorphisms of COX-2 -8473T/C were associated with the susceptibility to bladder cancer. The individ uals with the 8473C allele had a decreased risk of bladder cancer (OR=0.56, 95%CI=0. 35-0. 88). Conclusions Polymorphisrns of COX-2 -765G/C and -1195G/A are not associated with the susceptibility to bladder cancer. However, COX-2 -8473T/C can reduce the risk of bladder cancer.
