异丙酚联合电休克对抑郁大鼠海马和纹状体谷氨酸含量的影响 被引量:5

Effects of propofol combined with electroconvulsive therapy on glutamate content in hippocampus and corpus striatum in mentally depressed rats

摘要 目的探讨异丙酚联合电休克对抑郁大鼠海马和纹状体谷氨酸含量的影响。方法雄性成年SD大鼠60只,体重180~300g,随机分为5组(n=12):正常对照组(C组)、抑郁组(D组)、异丙酚组(P组)、电休克组(E组)和异丙酚联合电休克组(M组)。D组、P组、E组和M组建立21d动物不可预见慢性应激抑郁模型。造模后P组腹腔注射异丙酚100mg/kg,C组和D组给予等容量生理盐水,E组采用单纯电休克,M组腹腔注射异丙酚100mg/kg后进行电休克,各组隔天处理1次,连续2周共8次。于造模前1d、造模后第2天和处理结束后第2天,采用open-field法进行行为学评分;并进行液体消耗实验,计算糖水消耗比。行为学评分和液体消耗实验结束后第2天采用Morris水迷宫测试学习记忆功能。分离双侧海马和纹状体,计算谷氨酸含量。结果与C组比较,D组、P组、E组和M组造模后第2天和处理结束后第2天行为学评分和糖水消耗比降低,D组、P组和E组学习记忆功能减弱,D组和P组海马和纹状体谷氨酸含量升高,E组和M组海马和纹状体谷氨酸含量降低(P〈0.01);与D组比较,E组和M组处理结束后第2天行为学评分和糖水消耗比升高,海马和纹状体谷氨酸含量降低,E组学习功能减弱,M组学习记忆功能增强(P〈0.01);与E组比较,M组学习记忆功能增强,海马谷氨酸含量升高(P〈0.01)。结论异丙酚联合电休克通过恢复海马谷氨酸的含量,改善抑郁大鼠的学习记忆功能。 Objective Effects of propofol combined with electroconvulsive therapy (ECT) on glutamate content in hippocampus and corpus striatum in mentally depressed rats. Methods Sixty adult SD rats weighing 180- 300 g were randomly divided into 5 groups ( n = 12 each) : group Ⅰ control (C) ; group Ⅱ mental depression (D) ; group Ⅲ propofol (P); group Ⅳ electric shock (E) and groupV propofol + electric shock (M). In group D, P, E and M ( group Ⅱ-Ⅴ ) mental depression was induced by exposing the animals to different kinds of stresses every day for 21 days. After successful induction of mental depression, the animals received intraperitoneal propofol 100 mg/kg (in group P) or electric shock (in group E) or propofol 100 mg/kg + electric shock (in group M) every other day for 2 weeks. The establishment of mental depression was assessed by behavior using open-field test and glucose water consumption test (glucose water consumption/total fluid consumption). The two tests were performed one day before induction of mental depression, on the 2nd day after mental depression had been induced and on the 2nd day after treatment. The learning and memory functions were evaluated using Morris water maze on the 2nd day after treatment. The glutamate content in the hippocampus and corpus striatum was measured with chromatometry. Results The behavior scores and glucose water consumption were significantly decreased on the 2nd day after the establishment of mental depression and treatment in group D, P, E and M as compared with group C. The learning ability and memory of the animals in group D, P, and E were significantly decreased in group D, P and E as compared with group C and M. The glutamate content in hippoeampus and corpus striatum was significantly higher in group D and P and significantly lower in group E and M than in group C. Conclusion Propofol combined with electroconvulsive therapy can improve the learning ability and memory of mentally depressed rats through recovery of glutamate content in hippocamppus.
