摘要
基于电子延伸序列,克隆并分析了猪脂肪特异性蛋白27基因。各种组织提取总RNA,利用设计的引物进行RT-PCR,PCR产物与pMD19-T连接后转化E.coliJM109,检测阳性克隆并测序。猪FSP27基因的cDNA序列,其全长为745bp,开放阅读框为11-727bp,编码有238个氨基酸。同源分析结果表明,猪FSP27的核酸序列与人、小鼠和牛的同源性分别为86%、77.6%、82.3%,氨基酸序列的同源性分别为83.2%、74.4%、79.3%。组织分布结果显示:猪FSP27基因在多种组织均有分布。克隆的猪FSP27基因并注册GenBank(Accession.EU395789)。
The coding sequence of porcine fat specific protein27 gene were amplified based on the in silico sequence information in the present study. Total RNA was extracted from all kinds of organizations and mRNA sequences of gene were amplified by RT-PCR using designed primers. The PCR products were ligated into the pMD-19T vector, and then transformed into competent cells of E. coli JM109. Porcine fat specific protein27 gene cDNA sequence, the 745 bp in length, open reading frame was 11-727 bp detected by RT-PCR, encoding 238 amino acids residue protein. Homology analysis showed that porcine FSP27 and nucleic acid sequences human, mice and bovine were the homology of 86%, 77.6% and 82.3% amino acid sequence homology was 83.2%, 74.4%and 79.3%. The results showed that tissue distribution: Pig in a variety of organizations have FSP27 distribution. Cloning porcine FSP27 genes and registration GenBank (Accession.EU395789).
