摘要
目的使用光化学法构建食蟹猴局部脑缺血模型,为干细胞移植治疗提供必要的条件。方法选择8只健康成年食蟹猴,静脉注射35 mg/kg体重的玫瑰红B 5 min后用冷光源照射大脑皮层中央前回10 min。第一次注射后10 min钟注射第二次玫瑰红B,浓度与第一次相同。照射完毕后,向颞侧移动光源发光口,在第二点照射10min。利用影像学、神经功能缺损评分和组织病理学对模型进行评价。结果动物清醒后表现出不同程度的脑缺血症状与体征。病理学检查可见明显的坏死灶。坏死灶周围脑组织水肿,神经细胞变性坏死。结论成功的将光化学法首次应用于灵长类动物,为进行与临床有重要意义的神经细胞保护研究提供了重要的工具。
Objective To make a reproducible Photochemical Thrombosis animal model in Cynomolgus macaques and provide the necessary tool for treatment of cerebral infarction by transplantation of mesenchymal stem cells. Methods 8 male monkeys of 4 - 6 years were used in this study. Rose Bengal, a photosensitizing dye, was injected into the monkey via femoral vein (35 mg/kg) ,5 minutes later the cold lamp-house was focused on the precentral gyrus for 10 minutes. To evaluate the model by the method of histopathology, neurologic deficit score and PET. Results PET revealed the stable and formula cerebal ischemic injury in the fight cortex. Neurologic deficit score showed the neural symptom coincident. Histopathological examination showed that visible necrosis, hydrops and nerve cell degeneration necrosis in the place of damnification. Conclusion Photochemical Thrombosis animal model was first reproduced in Cynomolgus macaques. This procedure is rather economic and can produce large and stable infracted area. So it is very suitable for the nerve protection research in cerebral infarction.
