摘要
目的探讨我国三个特有的小型猪品系-中国农大小型猪,五指山小型猪,广西巴马小型猪脂联素受体1基因多态性分布,为这些小型猪在代谢性疾病和2型糖尿病研究中的应用提供基础资料。方法提取三个品系小型猪血液基因组DNA,针对脂联素受体1基因外显子5~内含子5的部分序列进行PCR扩增,对产物进行鉴定和测序,分析基因多态性。结果在所扩增的片段中共检测出5个sNPs——sNP1:G138A,发生在所有的小型猪品系中,并导致第45位的氨基酸由甘氨酸(Gly)转变为谷氨酸(Glu);SNP2:G185A,所有的纯合突变均发生在五指山小型猪品系中(4/6,66.67%),而所有的杂合突变则发生在中国农大小型猪品系中(3/10,30%);SNP3:G200A,全部发生在中国农大小型猪品系中,且均为杂合突变(3/10,30%);SNP4:C295T,发生在广西巴马小型猪品系中的突变既有杂合突变(1/6,16.67%)也有纯合突变(2/6,33.33%),发生在中国农大小型猪品系中的突变全为杂合突变(2/10,20%);SNP5:T337G,只出现在中国农大小型猪品系中,且为杂合突变(1/10,10%)。结论SNP1,在所有测定的小型猪品系中检出,这可能是所测小型猪共有的特性。SNP2,可能是小型猪品系之间的差异。SNP3和SNP4,可能是小型猪品系之间的差异,也可能是个体的差异。SNP5,可能是个体差异。
Objective To discuss polymorphism distribution of adiponectin receptor1 (AdipoR1) from three particular Chinese miniature pig strains--Nongda miniature pigs, Wuzhishan miniature pigs and Bama miniature pigs; Also, this essay supplies some fundamental information in the process of research for the application of metabolic diseases and type-2 diabetes mellitus on those miniature pigs. Methods The genomic DNA was extracted from three strains of Chinese native miniature pigs, part of sequence of AdipoR-1 gene exon5 was amplified by PCR. The amplified products were sequenced and identified, then the results of sequenced were compared with the result of NCBI. Results It was identified five SNPs--SNPI: G138A from the PCR amplification,which were discovered from all minipig strains and caused the change from Gly to Glu; SNP2: G185,A, it was identified that all homozygous mutations were discovered on Wuzhishan minipig strain and heterozygous mutations discovered from Zhongguonongda minipig strain; SNP3 : G200A, all for heterozygous mutations( 3/10,30% ) were discovered from Zhongguonongda minipig strain ; SNP4 : C295T was discovered from Bama mlnipig strain, both including heterozygous mutations ( 1/6,16.67 % ) and homozygous mutations(2/6,33.33% ), and discovered from Zhongguonongda minipig strain all for heterozygous mutation (2/10, 20% ); SNPS:T337G was only discovered from Zhongguonongda minipig strain and was heterozygous mutation (1/10, 10% ). Conclusion The discovery of SNP1 in all miniature pig strains, this may because the similar strait among the minipigs, The discovery of SNP2 was probably caused by the differences among the minipig strains. The discovery of SNP3 and SNP4 may have the same reason with SNP2, or from individual difference. The discovery of SNP5 was probably resulted in individual difference.
