Immunoregulatory Effect of Broken-BCG Mucous Membrane Immune Modifier BCG-b on Mice with Experimental Immune Hypofunction

摘要 目的观察卡介苗溶菌黏膜免疫调理剂(BCG-b)对实验性免疫功能低下小鼠的免疫调节作用。方法将BALB/c小鼠分为BCG-b高、中、低剂量组,每只分别隔日灌服BCG-b20、5和1U/0.2ml;阳性对照组每只隔日灌服阿胶液0.2ml,阴性对照组每只隔日灌服生理盐水0.2ml;正常对照组不给任何药物。除正常对照组外,其余各组均服药10次,并于首次服药后第14日皮下注射环磷酰胺,100mg/kg,建立免疫功能低下小鼠模型。以碳粒廓清法、血清溶血素测定法及3H-TdR掺入法分别检测各组小鼠单核-巨噬细胞吞噬功能、特异性体液免疫功能以及T淋巴细胞转化功能。结果与阴性对照组相比,BCG-b3个不同剂量组小鼠单核-巨噬细胞的吞噬功能和特异性抗体血清溶血素含量明显提高,小鼠T淋巴细胞转化功能呈双向性调节作用,即高、中剂量表现为抑制作用,低剂量表现为促进作用。结论BCG-b对实验性免疫功能低下小鼠具有明显的免疫调节作用。 Objective To observe the immunoregulatory effect of broken-BCG mucous membrane immune modifier BCG-b on the mice with experimental immune hypofunction. Methods Divide BALB/c mice into 6 groups. The mice in 3 test groups were administered every other day with 20, 5 and 1 U/0. 2 ml of BCG-b respectively, and those in positive and negative control groups with 0. 2 ml of bulk E-gelatin and 0. 2 ml of physiological saline respectively, for 10 times, and injected s.c. with cyclophosphamide to establish mouse model of immune hypofunction. However, the mice in normal control group were untreated. Determine the phagocytic function of mononuclear-macrophage by carbon particle clearance test, the specific humoral immune function by serum lysine assay, and the T lymphocyte transformation function by 3H-TdR incorporation method. Results Compared with those in negative control group, both the phagocytic function of mononuclear-macrophage and the specific serum lysine content of mice in 3 test groups increased significantly. However, the BCG-b at high and moderate dosages (20 and 5 U /0.2 ml) inhibited, while that at low dosage (1 U/0. 2 ml) promoted the T lymphocyte transformation function. Conclusion BCG-b showed significantly immunoregulatory effect on mice with experimental immune hypofunction.
